Photography with Nikon Microscope in Animal Facility
Film
ASA 50-400
- Black and white: T-Max 400
- Color slides: Ektachrome 400 or Fujichrome 400
- Stained sections: Fujichrome 50 works well
Procedure
- Set brightness control to 9 for daylight film, 8 for tungsten light, or 6 (or higher) for black and white.
- Spot meter reads 1% of area within double cross line
- Average (large circle) averages 30% of area
- Exposure adjust: 1/3 EV steps from -2 to -~
- +1 EV + extends shutter time
- – decreases shutter time
- -1 = about 1/2 exposure time
- -2 = about 1/4 exposure time
- For fluorescence microscopy with extremely dark background: 0 exposure adjustment if bright area of specimen covers area for spot meter; adjust if does not cover spot
- It is best to make slides and have prints made directly from slides if desired.
