Photography with Nikon Microscope in Animal Facility

Film

ASA 50-400

• Black and white: T-Max 400
• Color slides: Ektachrome 400 or Fujichrome 400
• Stained sections: Fujichrome 50 works well

Procedure

• Set brightness control to 9 for daylight film, 8 for tungsten light, or 6 (or higher) for black and white.
• Spot meter reads 1% of area within double cross line
• Average (large circle) averages 30% of area

• Exposure adjust: 1/3 EV steps from -2 to -~
  • +1 EV + extends shutter time
  • – decreases shutter time
    • -1 = about 1/2 exposure time
    • -2 = about 1/4 exposure time
• For fluorescence microscopy with extremely dark background: 0 exposure adjustment if bright area of specimen covers area for spot meter; adjust if does not cover spot
• It is best to make slides and have prints made directly from slides if desired.
